How to reconstitute peptides correctly

Peptides ship as a freeze-dried powder because they are far more stable dry than in solution. Before use that powder has to be dissolved, and this step is where most of the damage happens. Peptides are fragile molecules held together by weak bonds, and rough handling breaks them apart long before they ever reach you.

This guide covers the process itself, the arithmetic for working out a dose, and the errors that quietly ruin a vial.

What you need

  • Bacteriostatic water. Not sterile water, not tap water. Bacteriostatic water contains 0.9 percent benzyl alcohol, which suppresses bacterial growth and lets a vial stay usable for weeks after mixing. Plain sterile water has no preservative and gives you a single session before contamination becomes a real risk.
  • An insulin syringe for drawing and measuring, marked in units.
  • A larger syringe (3 ml or similar) for transferring the water, if the volume is more than an insulin syringe holds.
  • Alcohol pads for the rubber stoppers.
  • A place to store it. A fridge, not a freezer, and not the fridge door.

The method

  1. Let the vial reach room temperature. Taking a cold vial straight from the fridge and adding liquid causes condensation and thermal shock. Give it twenty minutes on the counter.
  2. Wipe both stoppers with an alcohol pad, the peptide vial and the bacteriostatic water vial, and let them dry.
  3. Draw your water. Decide the volume first (see the next section), then draw exactly that amount.
  4. Aim at the glass wall. Insert the needle at an angle and let the water run down the inside of the vial. Do not fire the stream directly onto the powder cake. The shear force from a jet of liquid hitting the powder is enough to break peptide bonds.
  5. Let it dissolve on its own. Most peptides go into solution within a minute or two without help. If some powder remains, roll the vial gently between your palms or swirl it.
  6. Never shake it. Shaking denatures peptides. If you see foam, you have already been too rough. A properly mixed vial is completely clear.
  7. Refrigerate. Store at 2 to 8 degrees Celsius, upright, away from light.

Working out the concentration

The volume of water you add is your choice. It does not change how much peptide is in the vial, only how concentrated the solution is. More water makes each unit on the syringe a smaller dose, which is easier to measure accurately.

The arithmetic is straightforward. An insulin syringe is marked in units, and 100 units is 1 ml.

Concentration = total peptide in the vial divided by millilitres of water added.

Take a 5 mg vial with 2 ml of bacteriostatic water added. That gives 5 mg across 200 units, so each unit holds 0.025 mg, or 25 micrograms. A 250 microgram dose is therefore 10 units.

Take the same 5 mg vial with 1 ml instead. Now 5 mg sits in 100 units, each unit is 50 micrograms, and the same 250 microgram dose is 5 units.

Both vials contain the same peptide. The second is simply harder to measure precisely, because a small error on the plunger is worth twice as much. For small doses, use more water.

How long it lasts once mixed

Reconstituted with bacteriostatic water and kept refrigerated, most peptides hold up for several weeks. The benzyl alcohol handles bacteria, but it does nothing about the slow chemical breakdown of the peptide itself, which continues regardless.

Practical guidance: mix what you will use within a month, keep it cold and dark, and do not carry vials around at room temperature. Heat and light are the two things that degrade peptides fastest after rough handling.

Mistakes worth avoiding

  • Shaking the vial. The single most common way to destroy a peptide. Swirl, never shake.
  • Using sterile water for a multi-dose vial. No preservative means no protection once the stopper has been pierced more than once.
  • Spraying water onto the cake. Run it down the glass instead.
  • Freezing a mixed vial. Freezing and thawing a solution damages peptides. Dry powder can be frozen; solution should not be.
  • Storing in the fridge door. The door is the warmest part of a fridge and swings in temperature every time it opens. Use a shelf at the back.
  • Assuming cloudy is normal. A correctly mixed vial is clear. Cloudiness, floating particles or anything that will not dissolve means something is wrong, and the vial should not be used.
